DATA SET FROM FUNCTIONAL DISTANCE MEDIATES PLANT INTERACTIONS

Dataset from experimets developed in the article "FUNCTIONAL DISTANCE MEDIATES PLANT INTERACTIONS". The outcome of plant interactions is strongly dependent on the set of physiological, morphological, and reproductive traits harboured by the interacting species. Two trait-based mechanisms are behind these plant interactions: a) trait dissimilarities, when species with similar traits compete more due to niche overlap, and b) trait hierarchies, when species with more efficient traits outcompete species with inefficient traits. Trait dissimilarities present a negative relationship between plant functional distance and interaction strength, and a positive relationship for trait hierarchies. We hypothesised that functional distance between species affect the outcome of plant interactions through both mechanisms. We established a manipulative experiment with ten species growing in hetero- and conspecific pairwise combinations and evaluated thirteen above- and belowground traits. We estimated neighbour effect by measuring the relative change (RC) in trait values of a focal species (RCfocal) caused by the presence of a heterospecific neighbour (RCneighbour), contrasted with the changes induced by a conspecific neighbour. We explored i) which traits were more affected by neighbours, ii) which species were more affected as focal or neighbour, and iii) how the neighbour effect varied with the functional distance between pairs. Neighbour effects on focal traits showed a variable intensity, contingent upon the specific identities of both plant species within the pair. Despite the high variability in neighbour effects, species that grew with a functionally similar neighbour exhibited larger traits than those growing with a dissimilar one due to the prevalence of trait hierarchies over trait dissimilarities. The heterogeneity found on the neighbour effects responded mainly to differences between above- and belowground traits, reinforcing the need to consider them both for a mechanistic comprehension of community dynamics. Trait hierarchies prevailed but trait dissimilarities were also detected on specific root traits, indicating that both mechanisms simultaneously determine the outcome of plant interactions., Peer reviewed

DATASET A SPLICE-ALTERING HOMOZYGOUS VARIANT IN COX18 CAUSES SEVERE SENSORY-MOTOR NEUROPATHY WITH OCULOFACIAL APRAXIA

Cytochrome-c oxidase (COX) is part of the mitochondrial complex IV (CIV). COX deficiency is usually associated with tRNA variants, and less frequently with variants in COX assembly factors. Mutations in COX subunits encoded by mitochondrial DNA and nuclear DNA are rare, likely because most of them are associated to very severe phenotypes with early lethality. COX18, an assembly factor of CIV, has long been analyzed as a potential cause of mitochondrial disease. To date, only one patient has been identified carrying a homozygous missense variant in COX18, associated with neonatal encephalo-cardiomyopathy and axonal sensory neuropathy. Here, we describe a 40-year-old patient, asymptomatic until 7 months of age, who presented with progressive muscle weakness resembling spinal muscle atrophy type-2, associated with oculofacial apraxia and dysarthric speech. Electrophysiology analysis highlighted a severe sensory-motor neuropathy. Muscle biopsy showed striking and diffuse decreases of COX staining and a substantial reduction of CIV activity. Muscle biopsy showed no ragged-red fibers, although ultrastructural mitochondrial alterations were evident. A novel homozygous variant (c.598G>A), located in the last nucleotide of exon 3, was detected in COX18 by whole-exome sequencing, which affected the splicing donor site, as demonstrated by cDNA-seq. The patient fibroblasts express a truncated form of COX18 (COX18Δ112-240) capable of assembling CIV and CIV-involving supercomplexes. However, CIV activity was decreased. COX18 full-length (COX18-fl) overexpression partially rescued CIV activity in the patient fibroblasts. The rescue of the null CIV activity in COX18-KO-HEK293 cells by overexpressing of COX18Δ112-240 was significantly lower than in cells with COX18-fl. In addition, cox-18 downregulation in C. elegans resulted in slow growth and, diminished reduced motility phenotypes and as well as severe fragmentation of the mitochondrial network. Our case expands the phenotypes associated with COX18 variants and supports the pathogenic role of COX18 as the cause of a severe encephaloneuropathy syndrome, The authors acknowledge the patient and his family, as well as the patient association ASENSE for all the support to this work. MECHANICAL AND FUNCTIONAL RESPONSES IN ASTROCYTES UNDER ALTERNATING DEFORMATION MODES USING MAGNETO-ACTIVE SUBSTRATES

This work introduces NeoMag, a system designed to enhance cell mechanics assays in substrate deformation studies. NeoMag uses multidomain magneto-active materials to mechanically actuate the substrate, transmitting reversible mechanical cues to cells. The system boasts full flexibility in alternating loading substrate deformation modes, seamlessly adapting to both upright and inverted microscopes. The multidomain substrates facilitate mechanobiology assays on 2D and 3D cultures. The integration of the system with nanoindenters allows for precise evaluation of cellular mechanical properties under varying substrate deformation modes. The system is used to study the impact of substrate deformation on astrocytes, simulating mechanical conditions akin to traumatic brain injury and ischemic stroke. The results reveal local heterogeneous changes in astrocyte stiffness, influenced by the orientation of subcellular regions relative to substrate strain. These stiffness variations, exceeding 50% in stiffening and softening, and local deformations significantly alter calcium dynamics. Furthermore, sustained deformations induce actin network reorganization and activate Piezo1 channels, leading to an initial increase followed by a long-term inhibition of calcium events. Conversely, fast and dynamic deformations transiently activate Piezo1 channels and disrupt the actin network, causing long-term cell softening. These findings unveil mechanical and functional alterations in astrocytes during substrate deformation, illustrating the multiple opportunities this technology offers., C. G.-C. and M.
