DATASET FOR ARTICLE RELATION BETWEEN HLB NUMBER AND PREDOMINANT DESTABILIZATION PROCESS FOR MICROFLUIDIZED NANOEMULSIONS FORMULATED WITH LEMON ESSENTIAL OIL [DATASET]

Lemon essential oil (LEO) is associated with a multitude of health benefits due to its anticancer, antioxidant, antiviral, anti-inflammatory and bactericidal properties. Its drawback is that it is very sensitive to oxidation by heat. For this reason, researchers are increasingly investigating the use of LEO in nanoemulsions. In this work, we used laser diffraction, rheology and multiple light scattering techniques to study the effects of different HLB numbers (indicating different mixtures of Tween 80 and Span 20) on the physical stability of nanoemulsions formulated with LEO. We found that different HLB numbers induced different destabilization mechanisms in these emulsions. An HLB number lower than 12 resulted in an Ostwald ripening effect; an HLB number higher than 12 resulted in coalescence. In addition, all the developed nanoemulsions exhibited Newtonian behavior, which could favor the mechanism of creaming. All emulsions exhibited not only a growth in droplet size, but also a creaming with aging time. These findings highlight the importance of selecting the right surfactant to stabilize nanoemulsions, with potential applications in the food industry

STUDY THE IMMUNOTOXIC EFFECT OF CYLINDROSPERMOPSIN [DATASET]

Cylindrospermopsin (CYN) is a cyanotoxin with an increasing occurrence, and therefore it is important to elucidate its toxicity profile. CYN has been classified as a cytotoxin, although the scientific literature has already revealed that it affects a wide range of organs and systems. However, research on its potential immunotoxicity is still limited. Thus, this study aimed to evaluate the impact of CYN on two human cell lines representative of the immune system: THP-1 (monocytes) and Jurkat (lymphocytes). CYN reduced cell viability, leading to mean effective concentrations (EC50 24 h) of 6.00± 1.04 µM and 5.20± 1.20 µM for THP-1 and Jurkat cells, respectively, and induced cell death mainly by apoptosis in both experimental models. Moreover, CYN decreased the differentiation of monocytes to macrophages after 48 h of exposure. In addition, an up-regulation of the mRNA expression of different cytokines, such as interleukin (IL) 2, IL-8, tumor necrosis factor-alpha (TNF-α) and interferon-gamma (INF-ɣ), was also observed mainly after 24 h exposure in both cell lines. However, only an increase in TNF-_ in THP-1 supernatants was observed by ELISA. Overall, these results suggest the immunomodulatory activity of CYN in vitro. Therefore, further research is required to evaluate the impact of CYN on the human immune system. Data was acquired for the study published in the article titled: Anti-obesogenic effect of lupin-derived protein hydrolysate through modulation of adiposopathy, insulin resistance and gut dysbiosis in a diet-induced obese mouse. This study examines the effects of a lupin protein hydrolysate (LPH) on obesity, adipose tissue dysfunction and gut dysbiosis in obese mice. LPH was characterized and protein concentration, ashes, moisture, fiber, fats, soluble sugars and phenol content were measured as described. Amino acid composition was determined using standard amino acid mix solution. The molecular weight profile was obtained by molecular exclusion chromatography. Eight-week-old male C57BL6/N mice were housed under standard conditions and mice were randomly assigned to three groups: mice fed a standard diet (SD), a high-fat diet (HFD) or an HFD and treated intragastrically with LPH (HFD+LPH) at a dose of 100mg/kg five days a week for 12 weeks. The SD and HFD groups were treated with vehicle under the same conditions as the HFD+LPH group. Food consumption and individual body weight were measured weekly. Animals were sacrificed with an intraperitoneal injection of sodium thiopental and blood was collected by cardiac puncture. Epidydimal adipose tissue (EpiWAT) was dissected, snap-frozen and stored at -80ºC until use or fixed in a 4% paraformaldehyde (PFA) solution. Stool was collected prior to euthanasia and stored at -80ºC. Serum biochemical profile was measured by chemoluminiscence in the Cobas Integra 400 (Roche Diagnostics, Indianapolis, IN, USA) at the Estación Biológica de Doñana (EBD-CSIC, Seville, Spain). Serum concentrations of leptin, adiponectin and insulin were quantified using enzyme-linked immunosorbent assays (ELISA) kits. Adipose tissue samples fixed in 4% PFA were dehydrated, embedded in paraffin blocks and sliced 4μm thick, stained with hematoxylin-eosin and cover mounted for posterior observation. Photomicrographs were obtained using Leica THUNDER microscope and Leica Application Suite X software. Adipocyte morphology and number of crown-like structures was blinded analyzed using ImageJ v1.53h public software. High throughput 16S rRNA gene amplicon analysis was performed using total DNA extracted from stool. 16S rRNA V3–V4 region was sequenced using MiSeq protocol and instruments. The raw sequences were processed using mothur., Dataset entitled “hydrolysate_composition.xlsx” includes macromolecular and aminoacidic composition (first sheet) and chromatogram information (second sheet) of LPH obtained from three independent measurements. Experimental data of copper leaching kinetics from non-pulverized printed circuit boards are compiled. They consist of the leached copper values as a function of time for the different tests performed. The copper concentrations were measured by atomic absorption spectrophotometry. Moreover, scanning electron microscope photomicrographs and energy dispersive X-ray spectroscopy data of a PCB sample has been included.
