SPLICING ASSAYS OF CHEK2 VARIANTS AND MICRODELETIONS LOCATED AT EXONS 8 AND 10: IDENTIFICATION OF SPLICING REGULATORY ELEMENTS

[Description of methods used for collection/generation of data] - Bioinformatics: DeepCLIP analysis (https://deepclip-web.compbio.sdu.dk/) - Sanger sequencing - Fluorescent Fragment Analysis., This dataset corresponds to a comprehensive study of the alternatively spliced exons 8 and 10 of the breast cancer susceptibility gene CHEK2. For this purpose, we have used a minigene with exons 6 to 10, where we introduced 12 internal microdeletions of exons 8 and 10 and 87 variants by site-directed mutagenesis that were assayed in MCF-7 cells. We identified three minimal (10,11,15-nt) regions essential for exon recognition. Thirty-eight variants impaired splicing, and 3 of which were classified as pathogenic/likely pathogenic variants according to ACMG/AMP (American College of Medical Genetics and Genomics/Association for Molecular Pathology)-based guidelines, so carrier patients and families may benefit from tailored prevention protocols and personalized therapies., EAV lab is supported by grants from the Spanish Ministry of Science and Innovation, Plan Nacional de I+D+I 2013-2016, ISCIII (PI20/00225) co-funded by FEDER from Regional Development European Funds (European Union). Comparison of the CoFeP-based electrodes for OER in 1 M KOH in chronological order., The development of high current density anodes for the oxygen evolution reaction (OER) is fundamental to manufacturing practical and reliable electrochemical cells. In this work, we have developed a bimetallic electrocatalyst based on cobalt–iron oxyhydroxide that shows outstanding performance for water oxidation. Such a catalyst is obtained from cobalt–iron phosphide nanorods that serve as sacrificial structures for the formation of a bimetallic oxyhydroxide through phosphorous loss concomitantly to oxygen/hydroxide incorporation. CoFeP nanorods are synthesized using a scalable method using triphenyl phosphite as a phosphorous precursor. They are deposited without the use of binders on nickel foam to enable fast electron transport, a highly effective surface area, and a high density of active sites. The morphological and chemical transformation of the CoFeP nanoparticles is analyzed and compared with the monometallic cobalt phosphide in alkaline media and under anodic potentials. The resulting bimetallic electrode presents a Tafel slope as low as 42 mV dec–1 and low overpotentials for OER. For the first time, an anion exchange membrane electrolysis device with an integrated CoFeP-based anode was tested at a high current density of 1 A cm–2, demonstrating excellent stability and Faradaic efficiency near 100%. 7 Pág., Update notice Author Correction: The GenTree Dendroecological Collection, tree-ring and wood density data from seven tree species across Europe (Scientific Data, (2020), 7, 1, (1), 10.1038/s41597-019-0340-y) Scientific Data, Volume 7, Issue 1, 1 December 2020, Article number 114, The dataset presented here was collected by the GenTree project (EU-Horizon 2020), which aims to improve the use of forest genetic resources across Europe by better understanding how trees adapt to their local environment. This dataset of individual tree-core characteristics including ring-width series and whole-core wood density was collected for seven ecologically and economically important European tree species: silver birch (Betula pendula), European beech (Fagus sylvatica), Norway spruce (Picea abies), European black poplar (Populus nigra), maritime pine (Pinus pinaster), Scots pine (Pinus sylvestris), and sessile oak (Quercus petraea). Tree-ring width measurements were obtained from 3600 trees in 142 populations and whole-core wood density was measured for 3098 trees in 125 populations. This dataset covers most of the geographical and climatic range occupied by the selected species. The potential use of it will be highly valuable for assessing ecological and evolutionary responses to environmental conditions as well as for model development and parameterization, to predict adaptability under climate change scenarios., This publication is part of a project that has received funding from the European Union’s Horizon 2020 research and innovation programme under grant agreement No. 676876 (GenTree).
