DIFFERENT GENE SETS ARE ASSOCIATED WITH AZACITIDINE RESPONSE IN VITRO AND MYELODYSPLASTIC SYNDROME PATIENTS

  • IJC, Admin
Myelodysplastic syndromes (MDS) are a heterogeneous group of hematopoietic disorders and characterized by dysplasia, ineffective hematopoiesis and predisposition to secondary acute myeloid leukemia (sAML). Azacitidine (AZA) is the standard care for high-risk MDS patients not eligible for allogenic bone marrow transplantation. However, only half of the patients respond and all patients eventually relapse. Response predicting biomarkers and combinatorial drugs targets enhancing response and its duration are needed. Here, we have taken a dual approach. First, we have evaluated genes encoding chromatin regulators for their capacity to modulate AZA response. We were able to validate several genes, whose genetic inhibition affected the cellular AZA response, including four genes encoding components of ISWI chromatin remodelling complex pointing towards a specific function and co-vulnerability. Second, we have used a classical cohort analysis approach measuring the association of expression of a gene panel in bone marrow samples collected at diagnosis of 36 MDS patients. The gene panel included the identified AZA modulators, genes known to be involved in AZA metabolism and previously identified candidate modulators. In addition to confirming a number of previously made observations, we were able to identify several new associations, such as SMARCA5 as positive indicator for AZA response, or NSUN3 that correlated with increased overall survival. Taken together we have identified a number of genes associated with AZA response in vitro and in patients. These groups of genes are largely non-overlapping suggesting that different gene sets need to be exploited for the development of combinatorial drug targets and response-predicting biomarkers.

SUPPLEMENTAL DATA AND CODE OF: IDENTIFYING FUNCTIONAL URBAN AREAS IN ECUADOR USING A VARYING TRAVEL TIME APPROACH

  • Obaco Álvarez, Moisés Lenyn
  • Royuela Mora, Vicente
Identifying integrated urban areas is an important issue for urban analysis and policy evaluation. In this paper, we provide a set of FUAs suitable for monitoring urbanization in Ecuador, a less developed country without commuting data. We do so substituting such socioeconomic flows by available information on road structure, which allow us to work with accessibility based on a varying travel time. In order to build Ecuadorean FUAs, we calibrate our methodology considering Colombian data. We perform several sensitivity analysis and robustness checks to Ecuador with alternative sources of socioeconomic flows. We conclude that the use of GIS data and the calibrated thresholds provide a set of FUAs close to the ones that would result of using commuting information.

SUPPLEMENTAL DATA OF: OPEN ACCESS AVAILABILITY OF CATALONIA RESEARCH OUTPUT: CASE ANALYSIS OF THE CERCA CENTRES

  • Rovira i Fernàndez, Anna
  • Abadal, Ernest
  • Urbano, Cristóbal
Sample of 3,730 journal articles published by authors from Catalonia's CERCA research centres between 2011 and 2015 and available on Web of Science (out of a total output of 44,423) identifying how many were available in open access, full-text format. The results revealed that 75,8% of the total (2,828 articles) had at least one version available in open access, but just 52% (1,940 articles) had at least one version available in either journals (whether pure or hybrid open access journals or those with embargo periods) or repositories, a finding that highlights the powerful role played by academic social networks in the sharp increase in open access availability. Of the 2,828 articles for which at least one open access version was found, a total of 9,868 copies were located. With respect to versions, the publisher’s final version, i.e. the type formatted for publication by journal publishers, was found in 75,3% of cases. The number of articles published in open access journals (567) was very close to the number of articles published in hybrid journals or journals with embargo periods (624). Only 40,4% of the articles in the sample were located in repositories, being the subject repositories the heaviest used. Fifty percent of the articles (1,881 publications) were posted on academic social networks, the most popular of which were ResearchGate and Academia. According to thematic areas, all six areas (science, life sciences, medical and health sciences, engineering and architecture and humanities) exceeded 70% of articles in open access.

SUPPLEMENTAL DATA OF: METABOLIC, MITOCHONDRIAL, RENAL AND HEPATIC SAFETY OF ENFUVIRTIDE AND RALTEGRAVIR ANTIRETROVIRAL ADMINISTRATION: RANDOMIZED CROSSOVER CLINICAL TRIAL IN HEALTHY VOLUNTEERS

  • Garrabou Tornos, Glòria
  • Martínez, Esteban
Anonymous data of two clinical trials. Twenty-six healthy participants were randomized to T20/90mg vs. placebo (n = 12) or RAL/400mg vs. placebo (n = 14) every 12h in two 7-day periods separated by a 4-week washout period. Major end-points were changes in lipid profile (total cholesterol, high-density-lipoprotein (HDL)-cholesterol, low-density-lipoprotein (LDL)-cholesterol, triglycerides), insulin resistance (glucose) and mitochondrial toxicity (mitochondrial DNA content–mtDNA–in peripheral blood mononuclear cells). Renal and hepatic toxicity (creatinine, alanine transaminase (AST), alanine aminotransferase (ALT), bilirubin and total plasma proteins) and overall safety were also analysed. Effect of period, treatment, and basal measures were evaluated for each end-point.

This dataset contains the information on our recent body of work on Ag-induced transformation of FeMn alloys and all the relevant data files. In this work, attempt to explain the austenite-martensite phase transformation occurring in FeMn alloys upon additions of Ag. In the study, a combined experimental and theoretical approach are applied to reveal the mechanism behind the phase transformation. While equiatomic FeMn and FeMn-1Ag alloys possess a fully austenitic structure, a change in the crystallographic structure is observed upon addition of 3 and 5 wt% of Ag, where a mixture of γ austenite and ε martensite phases is observed. Compression tests reveal that such structural transition causes an increase of the yield stress. The evolution of microstructure with the Ag content can be understood from theoretical calculations which show that Ag atoms prefer the intrinsic stacking fault (ISF) sites, revealing lower energy for the ε atomic plane sequence. This causes local depletion of the electronic charge, therefore weakening the interatomic bonds at the ISF plane and facilitating the phase transition. In addition, the total energy difference between the γ and ε phases decreases upon Ag addition. This enables the coexistence of both phases in the sample with 5 wt% Ag. Both experimental and theoretical data agree that the magnetization value gradually increases upon Ag addition. This is due to the local stress that is introduced by Ag atoms, which expand the Ag-Fe and Ag-Mn first neighbour interatomic bonds compared to FeMn. This stress results in electronic charge transfer that locally alters the Fe and Mn atomic magnetic moments.
