[Background]: Cancer is a global health problem accounting for nearly one in six deaths worldwide. Conventional treatments together with new therapies have increased survival to this devastating disease. However, the persistent challenges of treatment resistance and the limited therapeutic arsenal available for specific cancer types still make research in new therapeutic strategies an urgent need., [Methods]: Chloroquine was tested in combination with different drugs (Panobinostat, KU-57788 and NU-7026) in 8 human-derived cancer cells lines (colorectal: HCT116 and HT29; breast: MDA-MB-231 and HCC1937; glioblastoma: A-172 and LN-18; head and neck: CAL-33 and 32816). Drug´s effect on proliferation was tested by MTT assays and cell death was assessed by Anexin V-PI apoptosis assays. The presence of DNA double-strand breaks was analyzed by phospho-H2AX fluorescent staining. To measure homologous recombination efficiency the HR-GFP reporter was used, which allows flow cytometry-based detection of HR stimulated by I-SceI endonuclease-induced DSBs., [Results]: The combination of chloroquine with any of the drugs employed displayed potent synergistic effects on apoptosis induction, with particularly pronounced efficacy observed in glioblastoma and head and neck cancer cell lines. We found that chloroquine produced DNA double strand breaks that depended on reactive oxygen species formation, whereas Panobinostat inhibited DNA double-strand breaks repair by homologous recombination. Cell death caused by chloroquine/Panobinostat combination were significantly reduced by N-Acetylcysteine, a reactive oxygen species scavenger, underscoring the pivotal role of DSB generation in CQ/LBH-induced lethality. Based on these data, we also explored the combination of CQ with KU-57788 and NU-7026, two inhibitors of the other main DSB repair pathway, nonhomologous end joining (NHEJ), and again synergistic effects on apoptosis induction were observed., [Conclusion]: Our data provide a rationale for the clinical investigation of CQ in combination with DSB inhibitors for the treatment of different solid tumors., Peer reviewed

(2024, https://doi.org/10.20350/digitalCSIC/16077)., This dataset includes all raw observations (mostly in native instrument format) collected during the FaSt-SWOT oceanographic campaign (R/V SOCIB) carried out in the Balearic Sea (Western Mediterranean) between 25 April and 10 May 2023 (Leg 1 from 25 to 28 April and Leg 2 from 7 to 10 May). This experiment was part of the FaSt-SWOT project (PID2021-122417NB-I00 funded by MCIN/AEI/10.13039/501100011033/FEDER,UE), with the objectives to improve the characterization and understanding of small-scale ocean structures and contribute to SWOT satellite cal/val activities during its initial fast-sampling phase., Observations were collected both by ship-based instruments (CTD, Moving Vessel Profiler, thermosalinograph, ADCP and weather station) and autonomous platforms (surface drifters and gliders). They include measurements of both physical (temperature, salinity, currents) and biogeochemical (oxygen, chlorophyll, turbidity, photosynthetically active radiation) ocean variables., The data collection was mainly supported by the FaSt-SWOT project, funded by the Spanish Ministry of Science and Innovation, the Spanish Research Agency and the European Regional Development Fund (MCIN/AEI/10.13039/501100011033/FEDER, UE) under Grant Agreement PID2021-122417NB-I00. LANDSCAPE FEATURES INFLUENCING GENE FLOW AND CONNECTIVITY OF AN ENDANGERED PASSERINE [DATASET]

Within the dataset you can find the raster data (.asc) used for the optimization procedure to create resistance models for both years (1990/2018). Raster layers show the different landscape variables used in the study which were initially obtained from CORINE as it is explained in the manuscript. There is also a Genepop file containing the data related with microsatellites amplification (11 polymorphic microsatellites) for each individual at the different areas of study. Additional text files contains both matrix, genetic (Dps) and euclidean geographic (kilometers) distances between each pair of individuals sampled. With all this data it is possible to run resistanceGA analyses, as the ones explained in the article, in order to obtain the potential impact of the environment on gene flow. Some examples of studies related to this kind of analyses are: Flores-Manzanero et al., 2019 (https://doi.org/10.1002/ece3.4762) ; Peterman 2018 (https://doi.org/10.1111/2041-210X.12984; ([https://doi.org/10.1111/2041-210X.12984](https://doi.org/10.1111/2041-210X.12984)., Effective exchange of individuals among populations (i.e. gene flow) is one crucial aspect to maintain genetic diversity and viability of populations, especially in the case of threatened species. Landscape composition and structure may facilitate or limit individual movement within and among populations. We used a landscape genetics (LG) approach to assess the connectivity patterns of the threatened Dupont’s lark (Chersophilus duponti duponti), considering their genetic patterns and the landscape features associated with its gene flow in Spain. We analysed genetic structure and relatedness based on 11 species-specific polymorphic microsatellites on 416 Dupont’s lark individuals sampled across peninsular Spain between 2017 and 2019, covering most of the European distribution of the species. To assess the relationship between the landscape configuration and the species gene flow, we estimated genetic distance at the individual level (Dps). Next, we built a set of environmental surfaces from two time points (1990 and 2018), based on factors (land use and topography) influencing individuals’ movement. We then obtained resistance surfaces from an optimization process on landscape variables, and LG analyses were done for single and composite surface models for each year separately. Our findings from both time points demonstrate, for the first time, that scatter or mosaic structured vegetation composed by low agricultural and tree cover and high presence of sclerophyllous shrubs favoured Dupont’s lark dispersal, while dense and continuous tree cover, as well as areas of intensive agriculture, were limiting factors. Our results suggest the importance of steppe habitat patches both for the species establishment and dispersal. Besides, our results provide key information to develop specific conservation measures, including focusing on conserving and restoring steppe habitats as scattered and/or mosaic structured vegetation that could warrant the connectivity and persistence of Dupont’s lark populations., Peer reviewed
