THE HIV/AIDS VACCINE CANDIDATE MVA-B ADMINISTERED AS A SINGLE IMMUNOGEN IN HUMANS TRIGGERS ROBUST, POLYFUNCTIONAL, AND SELECTIVE EFFECTOR MEMORY T CELL RESPONSES TO HIV-1 ANTIGENS

Attenuated poxvirus vectors expressing human immunodeficiency virus type 1 (HIV-1) antigens are considered promising HIV/AIDS vaccine candidates. Here, we describe the nature of T cell immune responses induced in healthy volunteers participating in a phase I clinical trial in Spain after intramuscular administration of three doses of the recombinant MVA-B-expressing monomeric gp120 and the fused Gag-Pol-Nef (GPN) polyprotein of clade B. The majority (92.3%) of the volunteers immunized had a positive specific T cell response at any time postvaccination as detected by gamma interferon (IFN-γ) intracellular cytokine staining (ICS) assay. The CD4(+) T cell responses were predominantly Env directed, whereas the CD8(+) T cell responses were similarly distributed against Env, Gag, and GPN. The proportion of responders after two doses of MVA-B was similar to that obtained after the third dose of MVA-B vaccination, and the responses were sustained (84.6% at week 48). Vaccine-induced CD8(+) T cells to HIV-1 antigens after 1 year were polyfunctional and distributed mainly within the effector memory (TEM) and terminally differentiated effector memory (TEMRA) T cell populations. Antivector T cell responses were mostly induced by CD8(+) T cells, highly polyfunctional, and of TEMRA phenotype. These findings demonstrate that the poxvirus MVA-B vaccine candidate given alone is highly immunogenic, inducing broad, polyfunctional, and long-lasting CD4 and CD8 T cell responses to HIV-1 antigens, with preference for TEM. Thus, on the basis of the immune profile of MVA-B in humans, this immunogen can be considered a promising HIV/AIDS vaccine candidate., Peer reviewed

Inscripción que recorre en el friso de los muros de la capilla todo su contorno perimetral. Tiene su comienzo en el lado izquierdo del retablo mayor del presbiterio, recorriendo su primera parte el friso en madera y después pintada en negro sobre el muro en fondo blanco. La inscripción conmemora la fundación de la capilla en 1440 por don Juan Hurtado de Mendoza, Señor de Cañete. En el último tercio del siglo XVI fue reedificada por sus biznietos, Don Rodrigo de Mendoza, clavero de la Orden de Alcántara y Don Francisco de Mendoza, arcediano de Moya y Toledo, acontecimientos a los que alude también la inscripción. EFFECTS OF STSP6A OVEREXPRESSION AND SILENCING ON POTATO STOLONS

For each sample of the two comparisons (35S-StSP6A versus Wild Type and StSP6A-RNAi versus Wild Type) two biological replicates were prepared. In the case of 35S-StSP6A and StSP6A-RNAi each replicate was composed by a pool of several transgenic lines.-- Experiment type: Expression profiling by array., Comparisons (35S-StSP6A versus Wild Type and StSP6A-RNAi versus Wild Type., Summary StSP6A is a protein with sequence homology to the Arabidopsis FLOWERING LOCUS T (FT). FT is a main component of the long range flowering promoting signal or florigen that induces photoperiodic flowering under long days (LDs). In potato, short days (SDs) promote tuber induction, and wild subspecies like Solanum tuberosum ssp andigena (wild type line 7540) are strictly dependent on SDs to tuberize. StSP6A transcription is induced in leaves and stolons at early stages upon transfer to SD inductive conditions, priotr to visible solon swelling. Transgenic andigena lines over-expressing the StSP6A gene under the control of the 35S promoter are able to tuberize under LD non-inductive conditions, whereas silencing of this gene by means of an specific RNAi construct are strongly delayed in tuber induction in SD inductive conditions. TRANSCRIPTIONAL CHANGES DURING PHOTOPERIODIC TUBER INDUCTION IN SOLANUM TUBEROSUM SSP ANDIGENA

All collected stolons (LD; and 2d, 4d, 6d and 8d under SD) were processed as described. Hybridization scheme with a loop design was applied to compare one sample against the rest of the samples in the induction time course.-- Experiment type: Expression profiling by array., Expression profiling analyses of early stages of tuber induction, plants of Solanum tuberosum ssp andigena (7540)., For expression profiling analyses of early stages of tuber induction, plants of Solanum tuberosum ssp andigena (7540) were used. This wild subspecies is strictly dependent on photoperiod for tuberisation, such that short days (SD) inductive conditions are required in order to trigger tuber induction in the stolons. Andigena plants were grown in the greenhouse under LD non-inductive conditions until a 10-leaf stage. They were subsequently transferred to inductive SD conditions (8 h light/16 h dark), and sampled at 0, 2, 4, 6 and 8 days after transfer to SDs. Tuber swelling was visible approximately 6-8 days after transfer to inductive conditions.
