We have previously shown that an HIV vaccine regimen including three doses of HIV-modified vaccinia virus Ankara vector expressing HIV-1 antigens from clade B (MVA-B) was safe and elicited moderate and durable (1 year) T-cell and antibody responses in 75% and 95% of HIV-negative volunteers (n = 24), respectively (RISVAC02 study). Attenuated poxvirus vectors expressing human immunodeficiency virus type 1 (HIV-1) antigens are considered promising HIV/AIDS vaccine candidates. Here, we describe the nature of T cell immune responses induced in healthy volunteers participating in a phase I clinical trial in Spain after intramuscular administration of three doses of the recombinant MVA-B-expressing monomeric gp120 and the fused Gag-Pol-Nef (GPN) polyprotein of clade B. The majority (92.3%) of the volunteers immunized had a positive specific T cell response at any time postvaccination as detected by gamma interferon (IFN-γ) intracellular cytokine staining (ICS) assay. The CD4(+) T cell responses were predominantly Env directed, whereas the CD8(+) T cell responses were similarly distributed against Env, Gag, and GPN. The proportion of responders after two doses of MVA-B was similar to that obtained after the third dose of MVA-B vaccination, and the responses were sustained (84.6% at week 48). Vaccine-induced CD8(+) T cells to HIV-1 antigens after 1 year were polyfunctional and distributed mainly within the effector memory (TEM) and terminally differentiated effector memory (TEMRA) T cell populations. Antivector T cell responses were mostly induced by CD8(+) T cells, highly polyfunctional, and of TEMRA phenotype. These findings demonstrate that the poxvirus MVA-B vaccine candidate given alone is highly immunogenic, inducing broad, polyfunctional, and long-lasting CD4 and CD8 T cell responses to HIV-1 antigens, with preference for TEM. Thus, on the basis of the immune profile of MVA-B in humans, this immunogen can be considered a promising HIV/AIDS vaccine candidate., Peer reviewed

INTERPLAY OF DEEP-MARINE SEDIMENTARY PROCESSES WITH SEAFLOOR MORPHOLOGY OFFSHORE MADEIRA ISLAND (CENTRAL NE-ATLANTIC)

The deep-water sedimentary processes and morphological features offshore Madeira Island, located in the Central-NE Atlantic have been scantly studied. The analysis of new multibeam bathymetry, echo-sounder profiles and few multichannel seismic reflection profiles allowed us to identify the main geomorphologies, geomorphic processes and their interplay. Several types of features were identified below 3800 m water depth, shaped mainly by i) the interplay between northward-flowing Antarctic Bottom Water (AABW) and turbidity currents and ii) interaction of the AABW with oceanic reliefs and the Madeira lower slope. Subordinate and localized geomorphic processes consist of tectono-magmatic, slope instability, turbidity currents and fluid migration. The distribution of the morphological features defines three regional geomorphological sectors. Sector 1 represents a deep seafloor with its abyssal hills, basement highs and seamounts inherited from Early Cretaceous seafloor spreading. Sector 2 is exclusively shaped by turbidity current flows that formed channels and associated levees. Sector 3 presents a more complex morphology dominated by widespread depositional and erosional features formed by AABW circulation, and localized mixed contourite system developed by the interplay between the AABW circulation and WNW-ESE-flowing turbidite currents. The interaction of the AABW with abyssal hills, seamounts and basement ridges leads to the formation of several types of contourites: patch drifts, double-crest mounded bodies, and elongated, mounded and separated drifts. The patch drifts formed downstream of abyssal hills defining an previously unknown field of relatively small contourites. We suggest they may be a result of localized vortexes that formed when the AABW’s flow impinges these oceanic reliefs producingthe erosional scours that bound these features. The bottom currents in the area are known to be too weak (1–2 cm s− 1) to produce the patch drifts and scours. Therefore, we suggest that these features could be relics at present, having developed when the AABW was stronger than today, as during glacial/end of glacial stages., SUBVENT

CHANGES IN DISTRIBUTION OF PHYTOPLANKTON FUNCTIONAL GROUPS DISTRIBUTION, PIGMENT COMPOSITION AND THE REALIZED NICHE OF DINOPHYSIS ACUMINATA AT THE ONSET OF AN UPWELLING EVENT

Dinophysis acuminata is the major cause of lengthy harvesting bans in shellfish growing areas in European Atlantic waters. In North Western Iberian shelf waters, the growing season of D. acuminata lasts throughout the upwelling season (ca. from March to September). Once environmental conditions trigger initial population growth, abundance fluctuations are coupled to the event-scale dynamics of coastal upwelling-relaxation/downwelling cycles. The “ASIMUTH-Rias” cruise (17–21 June 2013) in the Galician Rias (Pontevedra and Vigo) and adjacent shelf, during a DSP outbreak, aimed to explore small-scale physical processes associated with late spring blooms (> 103 cells L-1) of D. acuminata. The cruise coincided with the initiation of an upwelling pulse following relaxation and deepening of a previously formed thin layer of centric colony-forming diatoms. In this work, a niche (sensu Hutchinson) approach based on an Outlying Mean Index (OMI) analysis was used to describe the realized niche of D. acuminata during transient conditions. The OMI analysis was applied to fine-scale measurements of physical properties, HPLC-derived pigment composition, and phytoplankton functional groups in cross-shelf transects and in a 36-h study at a fixed station. Although having a wide niche breath, the realized niche of D. acuminata (cell maximum 5 ×103 cells L-1 between 3 and 5 m) was characterized by outflowing warmer (15-16 °C) waters with high turbulence (? < 10-6 m2 s-3), high light intensity, and low nitrate concentrations. These results show that during a transient upwelling-initiation scenario, mid-shelf waters provided a more suitable habitat for D. acuminata and the accompanying dinoflagellate populations than waters inside the rías
