ILEGAL BLENDS BETWEEN OLIVE AND SOFT DEODORIZED OILS (2021)

  • Moreda, Wenceslao
Excel file consisting of 14 sheets in which the calculation of the total diacylglycerol (DAG) content of a number of defective, non-defective, and soft deodorized olive oil samples, and of some of their corresponding mixtures, are given as examples. Data also show free acidity values and the theoretical DAG composition, together with the calculation of the R1 and R2 factors. It also includes the three resulting tables published as part of Food Chemistry, volume 330, 127226. Data must be read and interpreted within the context of such publication. Original chromatograms are not given since they are considered confidential., These data were generated in our lab, with the help of several laboratory assistants and master students, within the OLEUM project (http://www.oleumproject.eu): ‘Advanced solutions for assuring the authenticity and quality of olive oil at a global scale’ . The general objectives of the Project focus on olive oil fraud detection in a way that it improves the existing analytical methods and it has developed new strategies of analysis. It has been organized in seven work packages distributed in a number of tasks. This work is within Work Package 4 (‘Analytical solution addressing olive oil authentication issues’), and it concentrates on the detection of illegal blends, i.e. of illicit processing (deodorization) in extra virgin olive oil (EVOO) In our endeavor to detect soft deodorized olive oil in EVOO we have focused on the convenience of using two new factors (R1 and R2) obtained as a result of combining the total DAG content and the free acidity value of the samples under the microscope. We chose this methodology following our trend of using well-known, widely reputable routine parameters, avoiding in this way more complicated strategies that, although commonly used in the field of olive oil authentication, could require a more specific personnel training and laboratory apparatus. According to this, we demonstrated that since there is a relationship between free acidity and DAG concentration (both of them come from triacylglyceride hydrolysis and/or biosynthesis), and that such relationship breaks once the oil has gone through a refining process (free fatty acids are removed during the deodorization stage), it was possible to detect the presence of soft deodorized oil in EVOO by using a mathematical combination of both measurements at least to a certain extent. We actually proposed two factors to confirm the absence of soft deodorized oils in EVOO: R1 (10 x free acidity/DAGexp) >/= 0.23 and R2 (DAGexp-DAGtheor) < 0, in genuine EVOO, and we established that such approach was useful to detect the presence of soft deodorized olive oil when this was at least at 30 % in the mixture (Food Chemistry, volume 330, 2020, 127226). Samples were provided by Fera (Fera Science Ltd, Sand Hutton, York) and Institut des Corps Gras (ITERG, Canéjan, France)., The Project has received funding from the European Commission within the Horizon 2020 Program (2014–2020), GA no. 635690., Peer reviewed

  • Gómez, Pedro
Aphid borne viruses are responsible for major plant diseases, we monitored for three consecutive (2018-2020) seasons, the presence of aphid-borne viruses from a total of 292 samples of watermelon and squash that showed yellowing symptoms. The samples were collected from three major cucurbit-producing areas (Castilla La-Mancha, Alicante and Murcia) in Spain. We found that cucurbit aphid-borne yellow virus (CABYV) was the most common virus found (29%) in the plants from both crops. Likewise, except for squash samples from Castilla La-Mancha and Alicante, watermelon mosaic virus (WMV) was also found (23%) with a relatively high frequency in watermelon plants, while the occurrences of PRSV and ZYMV were low (<3%), and even CMV was undetected in both plant species. Additionally, two CABYV cDNA infectious clones (CABYV LP-63 and CABYV-MEC 12.1) were constructed for further characterization, with the LP63 isolate causing more severe symptoms and showing higher RNA accumulation than MEC12.1 in five cucurbits plant species., Funding: AGL2017-89550-R from the Agencia Estatal de Investigación (AEI) and FEDER (EU) funds., Peer reviewed

FASTA SEQUENCES FOR THE DROSOPHILA MELANOGASTER MANUALLY CURATED TRANSPOSABLE ELEMENTS (MCTE) LIBRARY

  • Rech, Gabriel E.
The compressed file contains two files: MCTE.fasta with the consensus TE sequences in fasta format and the MCTE_info.xlsx Excel file with information for every sequence in the MCTE.fasta file. We used the REPET package (v.2.5) (Flutre et al. 2011; Hoede et al. 2014; Quesneville et al. 2005) for performing TE annotations using a manually curated TE (MCTE) library of consensus sequences. Briefly, REPET is composed of two main pipelines, TEdenovo dedicated to de novo detection of TE families and TEannot for the annotation and analysis of TEs in genomic sequences. For the creation of the MCTE library, we first run the REPET (v.2.5) (Flutre et al. 2011; Hoede et al. 2014; Quesneville et al. 2005) TEdenovo pipeline with default parameters on 13 genomes Drosophila melanogaster natural strains. The manual curation of the identified consensuses involved three main procedures: removal of redundant sequences, the manual identification of potentially artifactual sequences and the classification of consensuses into families. Redundant sequences (consensus sequences present in more than one genome) were identified by first running the PASTEClassifier module from PASTEC with default options (Hoede et al. 2014). We also performed similarity clustering, multiple sequence alignments (MSA) of the clusters and generated consensus sequences for each MSA in order to obtain a consensus sequence representative of all the genomes. We manually explored the consensus sequences and their copies using the plotCoverage tool from REPET and discarded consensuses showing mainly a high number of small copies. The assignation of the consensus sequences into families was performed using BLAT (Kent 2002) against the curated canonical sequences of Drosophila TEs from the Berkeley Drosophila Genome Project (BDGP) v.9.4.1 (https://fruitfly.org/p_disrupt/TE.html). When no matches were found, we used RepeatMasker (v.4) (Smit 2015) with the release RepBaseRepeatMaskerEdition-20181026 of the RepBase (Bao et al. 2015)., Manually Curated Transposable Elements (MCTE) library from Drosophila melanogaster. De novo reconstruction was performed using TEdenovo pipeline from REPET package (v.2.5) (Flutre et al. 2011; Hoede et al. 2014; Quesneville et al. 2005). De-novo genome assemblies were sequenced using long-read sequencing technologies (Oxford Nanopore Technologies or Pacific Bioscience)., This file was created in the framework of the ERC grant number H2020-ERC-2014-CoG-647900, funded by the European Commission (https://erc.europa.eu/)., Peer reviewed
