Coastal marine environments — including lagoons, fjords, and estuaries — are characterized by pronounced environmental variability, with fluctuations in salinity, temperature, and nutrient levels shaping microbial community structure and function. To explore these dynamic systems, we sampled 26 sites between December 2021 and January 2023 along the coasts of France and Chile, spanning lagoons, estuaries, fjords, harbours, and both coastal and offshore marine environments. Surface waters were collected using either Niskin bottles or polycarbonate carboys, with additional deeper layers obtained at three sites. Monthly sampling at six French sites enables the assessment of seasonal dynamics. The presented dataset captures multiple environmental gradients, ranging from terrestrial–marine transitions and salinity fluctuations to eutrophication level, seasonal, geographic and disturbance-related variation. A total of 116 samples were analysed for 13 environmental parameters alongside metabarcoding and metagenomic sequencing. Temperature and salinity were recorded in the field immediately after sampling. Chlorophyll a was quantified fluorometrically following acetone extraction (Holm-Hansen et al. 1965; Lorenzen 1967). Dissolved organic carbon (DOC) was measured using high-temperature catalytic oxidation (Benner and Strom 1993) with a TOC analyser. Fluorescent dissolved organic matter (fDOM) peaks T and C (as proxy for protein like and humic like DOM, respectively) were quantified with a Jasco FP-8500 spectrofluorometer (Coble 1996). Leucine incorporation rates were determined via the centrifugation method (Smith and Azam 1992), and respiration rates were estimated via oxygen consumption in 5 ml glass vials using PreSens OxoDishes and a SensorDish® Reader. Inorganic nutrients were analyzed using a QuAAtro39 or AA3HR autoanalyzer (Grasshoff et al. 2009; Aminot and Kérouel 2007), Centre National de la Recherche Scientifique (CNRS), grant/award no. Water samples were collected from 13 sites along the French coast in the Gulf of Lyon and 13 sites along the Chilean Coast, spanning from Valparaiso to northern Patagonia and covering a wide range of coastal habitats. All 116 samples were processed for both metabarcoding and metagenomic library preparation. Metabarcoding of the 16S rRNA V4-V5 gene region yielded data for 115 samples (one failed library) comprising 53,392 amplicon sequence variants (ASVs) with some samples sharing as few as three ASVs. Metagenomic sequencing from all 116 samples enabled the reconstruction of 1372 medium to high-quality prokaryotic metagenome-assembled genomes (MAGs) following the standard of minimum information about a metagenome-assembled genome (MIMAG). The dataset also includes a gene catalogue of prokaryotic and eukaryotic genes exhibiting 31% of functional novelty. In addition, comprehensive environmental data were collected, including salinity, temperature, chlorophyll-a, cell densities, phosphate, nitrate, nitrite, ammonium, dissolved organic carbon (DOC), humic-like fluorescent dissolved organic matter (fDOM), protein-like fDOM, respiration and leucine incorporation rates.
Here, the ASV tables, all reconstructed MAGs, the gene catalogue of prokaryotic and eukaryotic genes, as well as the data files used as input for the technical validation presented in the associated data paper are posted. The file structure follows the structure of the Repository linked below. To avoid duplicated input files, the files were only uploaded to the directory of the scripts that the files were used in first. A copy of release version 1.0.0 of the repository is included in this data record (iowseq000048_gesifus.field.zip)., The study was financed by the German Science Foundation (DFG, BE 5937/2-3) and further financially supported via a RCPJANAH - Dotation (CPJ) from the University of Clermont Auvergne. Sampling activities in Chile were financially supported by the Chilean National Agency for Research and Development (ANID) through the COPAS Coastal project (FB210021), and through the Fondecyt 1211977 research project as well as the CNRS Laboratoire international associé program (IRP MAST CNRS INSU INSB INEE). Further funding was provided via Ecole Normale Supérieure de Paris, the Fondation de la Mer, the Institut de l’Océan via the Alliance Sorbonne Université, the Erasmus+ Programme of the European Union, This work contributes to the Institut de Ciències del Mar "Severo Ochoa Centre of Excellence" accreditation CEX2024-001494-S funded by AEI 10.13039/501100011033 of the Spanish Ministry of Science and Innovation, Peer reviewed

This dataset contains bulk RNA-seq data from GFP⁺ NG2⁺ or GFAP⁺ cells isolated by FACS following StarTrack labeling by in utero electroporation (IUE) at embryonic day E12, with cortical tissue dissociated at E14. A total of 10 samples are included (5 NG2⁺ and 5 GFAP⁺ biological replicates).
An Excel file (data_counts_fpkm) is provided containing gene-level raw read counts (*_count) and normalized expression values (*_fpkm) for each replicate. The file also includes differential expression analysis results for NG2 vs GFAP and GFAP vs NG2 comparisons (log2 fold change, p-values, and adjusted p-values), as well as gene annotations including gene name, chromosomal location, strand, gene length, biotype, functional description, and transcription factor family.
Raw sequencing data are provided as paired-end compressed FASTQ files (.fq.gz), where _1 and _2 correspond to forward (R1) and reverse (R2) reads from the same biological sample., How distinct cortical progenitor types and their temporal differences influence neuronal and glial development remains a key question in neurobiology. While Radial Glial Cells (RGCs) are well-established as Neural Progenitors (NPCs), NG2+NPCs have emerged as contributors with unique characteristics. Here, we combined in utero electroporation, lineage tracing and transcriptomic profiling to compare lineage outputs of NG2+ and GFAP+NPCs in the developing mouse cortex. Both generate neurons and glia, but with partially distinct patterns in lineage output, spatial distribution, and fate bias. NG2+NPCs undergo an early neurogenic phase followed by glial differentiation, producing glia with laminar distribution biases. In contrast, GFAP+NPCs sustain neurogenesis longer, producing a broader range of neurons and more constant glia during development. Our findings highlight the contribution of progenitor identity and temporal differences to distinct, partially overlapping lineage trajectories, supporting a model in which fate potential and timing co-regulate the spatial and laminar organization of cortical cell types., Ministerio de Ciencia, Innovación y Universidades

BLOOD DNA METHYLATION OF 1 MONTH-OLD MERINO EWE LAMBS (F1) BORN FROM DAMS (F0) SUPPLIED BETANE DURING GESTION [DATASET]
- Andrés, Sonia
- Giráldez, Francisco Javier
Twenty-eight pregnant Merino ewes were used to study the effect of including 4 g/kg of betaine in a complete pelleted ration on feed intake and animal performance of both, dams (F0) and female offspring (F1), including the first lactation period. To this end, fourteen ewes (F0-CTL group) received a complete pelleted ration (control diet) ad libitum during the last two months of gestation. The remaining animals (F0-BET group; n = 14) received the same diet, but with the addition of 4 g/kg of betaine. The results of the present study revealed that betaine supplementation during the gestation period of Merino ewes caused differentially methylated regions (DMRs) in the female offspring that were assessed at 53,649 and 2164 sites using MethylKit (q value < 0.05) and CGmap Tools (FDR < 0.05), respectively., This work was funded by Ministerio de Ciencia e Innovación (PID2021-126489OB-I00, MCIN/AEI/10.13039/501100011033, “FEDER, Una manera de hacer Europa”); ACRONYM: NUPROVI

Physical enrichment improves fish welfare and reduces negative traits in captivity. Solea senegalensis, a key species in European aquaculture, is often raised in intensive fiberglass tanks, which can lead to undesirable traits, particularly for future breeders. This study assessed the long-term effects of using a sand substrate during the on-growing phase on growth, cognition, and maturation in Senegalese sole females. Six tanks with juvenile soles were divided into two environments (three fiberglass, three sand), and sampled every six months for two years (n = 30 per group). This study measured biometrics, blood, gonad histology, and gene expression in the brain and gonad. Females from sand grew faster than females from fiberglass. The histological results showed that the sand group was more mature than the fiberglass group from the second sampling, where the sand group reached previtellogenic oocytes. Sex steroids and Lh levels were also significantly higher in the sand group after two years. These results in maturation were supported by the fhra and pgrexpression in the brain and gonads. Females from sand showed higher cognitive capacity according to c-fos, nr4a2, and bdnf expression. These results show that using sand as an enrichment helps improve growth, cognition, and reproduction in Senegalese sole females, This study received Portuguese national funds from FCT—Foundation for Science and Technology through projects UIDB/04326/2020 (DOI:10.54499/UIDB/04326/2020), UIDP/04326/2020 (DOI:10.54499/UIDP/04326/2020) and LA/P/0101/2020 (DOI:10.54499/LA/P/0101/2020) and from the Blue Bioeconomy Pact with the project Algae Vertical Nº C6449 | 5664-00000026 funded by the Portuguese Plan of Recovery and Resilience (PRR) and the European Union. In neurones, like in any other cell, their function often relies on the fine-tuning of their protein levels, which is achieved by the balance between protein synthesis and turnover. Defects in protein homeostasis frequently lead to neuronal dysfunction and neurological disorders. Given their extreme morphological complexity and high compartmentalization, neurones highly depend on the asymmetrical distribution of their proteome. The common belief is that proteins that sustain axonal, dendritic and synaptic functions are synthesized in the soma and then transported to distal neuronal compartments. However, there is a complementary mechanism by which the mRNAs, and not the proteins, are transported to distal subneuronal domains, and once they reach their destination, they are locally translated. Although once considered heretical, local translation (or local protein synthesis) is now widely accepted by the scientific community. Nonetheless, there is one question that remains largely unexplored in the field, and that is whether local translation in dendrites, axons and synapses is fully regulated by the neurone itself or if non-neuronal cells (e.g., glia) can modulate this mechanism in a non-cell-autonomous manner. Here, we combined primary neuronal cultures, astrocyte-derived extracellular vesicle (EVs) isolation, and proteomics to investigate whether astroglial EVs modulate local translation in axons. We show that EVs released by astrocytes exposed to amyloid-β peptide (Aβ) enhance protein synthesis specifically in distal axons and increase synaptic integrity. Proteomics analysis and western blotting identified the ribosomal protein Rps6 as an astroglial Aβ-EV cargo delivered to axons. Interestingly, genetic downregulation revealed the contribution of vesicular Rps6 to translation regulation in axons and synaptic integrity. To our knowledge, this is the first report that directly demonstrates glial control of local translation in neurones through EVs, revealing a novel glia-to-neurone communication mechanism in an experimental model of Alzheimer's disease (AD)., This work was supported by grants SAF2016-76347-R and PID2022-139451OB-I00 funded by MICIU/AEI/10.13039/501100011033 and by“ERDF/EU” (to JB), grants AARG-19-618303 and AARG-19-618303-RAPID from the Alzheimer´s Association (to JB), grant 2024RTE00652727from the Basque Department of Health (to JB), a start-up grant from theRamón y Cajal program (RYC-2016-19837 to JB) and start-up funds fromthe Basque Foundation for Science (IKERBASQUE to JB). SLAM (Simultaneous Localization and Mapping) is an efficient method for robot to percept surrendings and make decisions, especially for robots in agricultural scenarios. Perception and path planning in an automatic way is crucial for precision agriculture. However, there are limited public datasets to implement and develop robotic algorithms for agricultural environments. Therefore, we collected dataset “GrapeSLAM”. The ‘‘GrapeSLAM'' dataset comprises video data collected from vineyards to support agricultural robotics research. Data collection involved two primary methods: (1) unmanned aerial vehicle (UAV) for capturing videos under different illumination conditions, and (2) trajectories of the UAV during each flight collected by RTK and IMU. The UAV used was Phantom 4 RTK, equipped with a high resolution camera, flying at around 1 to 3 meters above ground level., The authors acknowledge the support of the Wageningen University, Chinese Academy of Agricultureal Sciences joint Ph.D. program.
