Besides the regulation of metabolic pathways, alternative electron transport is subjected to thiol-dependent regulation. We investigated the regulation of O2 reduction at photosystem I. The level of O2 reduction in leaves and isolated thylakoid membranes depends on the photoperiod in which plants are grown. We used a set of Arabidopsis (Arabidopsis thaliana) mutant plants affected in the stromal, membrane, and lumenal thiol network to study the redox protein partners involved in regulating O2 reduction. Light-dependent O2 reduction was determined in leaves and thylakoids of plants grown in short-day and long-day conditions using a spin-trapping electron paramagnetic resonance assay. In wild-type samples from short-day conditions, reactive oxygen species generation was double that of samples from long-day conditions, while this difference was abolished in several redoxin mutants. An in vitro reconstitution assay showed that thioredoxin m, NADPH-thioredoxin reductase C, and NADPH are required for high O2-reduction levels in thylakoids from plants grown in long-day conditions. Using isolated photosystem I, we also showed that reduction of a photosystem I protein is responsible for the increase in O2 reduction. Furthermore, differences in the membrane localization of m-type thioredoxins and 2-Cys peroxiredoxin were detected between thylakoids of short-day and long-day plants. Overall, we propose a model of redox regulation of O2 reduction according to the reduction power of the stroma and the ability of different thiol-containing proteins to form a network of redox interactions., We would like to thank Patrice Hamel (Ohio State University, USA) for scientific discussions and for sending us the seeds of the lto1 mutants. The Zur protein is a key player in coordinating this response in most species. Comparative proteomics conducted on the cyanobacterium Anabaena highlighted the more abundant proteins in a zur mutant compared to the wild type. Experimental evidence showed that the exoprotein ZepA mediates zinc uptake. Genomic context of the zepA gene and protein structure prediction provided additional insights on the regulation and putative function of ZepA homologs. Phylogenetic analysis suggests that ZepA represents a primordial system for zinc acquisition that has been conserved for billions of years in a handful of species from distant bacterial lineages. Furthermore, these results show that Zur may have been one of the first regulators of the FUR family to evolve, consistent with the scarcity of zinc in the ecosystems of the Archean eon., We are indebted to Rocío Rodríguez from the Proteomics Facility of IBVF for excellent assistance with sample preparation for mass spectrometry analyses and the analysis of proteomics data. 1) The file ordered_genomicPrediction_2445_99ind.vcf contains reference genotypes of 99 clones from the Spanish breeding programme: the consensus genotype of 15 registered clones and genotypes of 84 unregistered clones, genotyped with 2245 SNP markers. 2) File B23_phenotypes.xlsx contains phenotypes used in the study., Stone pine (Pinus pinea L.) is an emblematic tree species within the Mediterranean basin, with high ecological and economic relevance due to the production of edible nuts. Breeding programmes to improve pine nut production started decades ago in Southern Europe but have been hindered by the near absence of polymorphisms in the species genome and the lack of suitable genomic tools. In this study, we assessed new stone pine’s genomic resources and their utilisation in breeding and sustainable use, by using a commercial SNP-array (5,671 SNPs). Firstly, we confirmed the accurate clonal identification and identity check of 99 clones from the Spanish breeding programme. Secondly, we successfully estimated genomic relationships in clonal collections, an information needed for low-input breeding and genomic prediction. Thirdly, we applied this information to genomic prediction for total number of cones unspoiled by pests and their weight measured in three Spanish clonal tests. Genomic prediction accuracy depends on the trait under consideration and possibly on the number of genotypes included in the test. Predictive ability (ry) was significant for the mean cone weight measured in the three clonal tests, while solely significant for the number of cones in one clonal test. The combination of a new SNP-array together with the phenotyping of relevant commercial traits into genomic prediction models, proved to be very promising to identify superior clones for cone weight. This approach opens new perspectives for early selection., Peer reviewed
