GEOMETRIC MORPHOMETRIC CHARACTERIZATION OF THE BALKAN ALPINE GRASSHOPPER GENUS OROPODISMA UVAROV, 1942 (ORTHOPTERA, ACRIDIDAE, MELANOPLINAE), WITH DESCRIPTION OF TWO NEW SPECIES

  • Trillo, Marina
  • Ortego, Joaquín
In this study we employ a geometric morphometric approach to quantify morphological differences among taxa of the genus Oropodisma Uvarov, a complex of alpine grasshoppers that includes several narrow endemic species distributed across different mountain ranges of the Balkan Peninsula. The genus was described by Uvarov in 1942 and currently comprises ten recognized species, primarily distinguished by subtle differences in the shape of the phallus apex and furculae of males. Some populations from central Greece exhibit an uncertain taxonomic status, suggesting the possible existence of undescribed species. Quantitative morphological comparisons with specimens collected from the type localities of all currently recognized taxa revealed the presence of two new species in the Pindus Range, Greece: O. tzoumerkae sp. nov. from Mt. Tzoumerka and O. agrafae sp. nov. from Mt. Agrafa. Geometric morphometric analyses indicate that the taxa O. tymphrestosi and O. willemsei, distributed in adjacent mountain ranges from central Greece, do not show statistically significant differences in either of the two studied traits. This finding raises doubts about their taxonomic status but may also reflect the limitations of geometric morphometric approaches in capturing subtle differences in the complex structures that characterize internal male genitalia. Overall, our analyses highlight the importance of quantitative approaches in formally reassessing the taxonomic status of species complexes. The description of two new and geographically restricted species of Oropodisma further emphasizes the role of Mediterranean mountains as key centers of microgeographic speciation for alpine biotas., TED2021-129328B-I00; PID2021-123298NB-I00, Peer reviewed

CONTRIBUTION OF DNA METABARCODING TO THE ENVIRONMENTAL FUNGAL ASSESSMENTS IN HOSPITALS

  • Garcia-Gutierrez, Laura

  • Martín Sánchez, Pedro Mª
[Notes] This dataset ("Mycospitalomycs_metabarcoding") includes all revevant files and R scripts to reproduce: 1) Data preparation - from the original OTU matrix, including technical replicates and control samples, to the filtered fungal rarefied matrix; including the following these steps: (i) filtering Fungi, (ii) assessing the negative controls, (iii) assessing the mock community samples, (iv) assessing technical replicates, (v) plot OTUs vs Reads, (vi) rarefying the matrix, and (vii) preliminary NMDS ordination to find outliers. 2) Statistical analyses: (i) alpha diversity - fungal richness and other indices, (ii) beta diversity - NMDS ordination plots, (iii) PERMANOVA analyses to calculate the contribution of the key variables to explain the compositional variation of the mycobiomes, (iv) taxonomical descriptions, (v) Venn diagrams showig the indoor OTUs shared between hospitals and zones, (vi) qPCR data analysis, (vii) correlation matrix comparing variables related to eDNA, culturing and particle data, and (viii) indicator species associated with sample types and hospital zones. The statistical-analyses R script reproduces all figures and tables showing results from this study. Some relevant output tables have been included in this dataset. All dataset contents are detailed in the ReadMe file: "README_Mycospitalomics_metabarcoding", as well as other relevant information., [Background:] Hospitals are particularly sensitive environments where immunosuppressed patients might acquire invasive fungal infections (IFI). Therefore, it is necessary to carry out periodical environmental microbiological assessments that evaluate the fungal bioburden in air and surfaces from different hospital zones. Current microbiological monitoring protocols at healthcare settings are mostly based on cultivation, while environmental DNA (eDNA) assessments are still scarce and should be further evaluated. To fill this gap, this study combines a large sampling scheme, comprising > 200 samples (air, surface, dust and soil) collected from four zones at three Spanish hospitals in two campaigns (winter and autumn), with two eDNA approaches (DNA metabarcoding and quantitative PCR) to characterize the hospital mycobiomes (diversity, community composition and airborne load), compared to a parallel culture-dependent study., [Results:] Fungal richness was significantly higher in soil and air samples compared to indoor surface samples (vents and high-touch surfaces), as well as in samples collected in winter compared those taken in autumn. Intensive care units showed lower fungal richness compared to regular patient rooms, waiting rooms and entrance halls. The most important explanatory factors for the variance in community composition were the hospital and zone where samples were collected, the type of sample, and the sampling campaign., Hospital mycobiomes, represented by 1,900 OTUs, were affiliated to 4 phyla (mostly Ascomycota - 53 % and Basidiomycota - 41.3 %), 35 classes, 114 orders, 305 families, 643 genera, and 535 species. The dominant genera, in both air and surfaces samples from the three hospitals, were Cladosporium Alternaria, Aureobasidium, Penicillium, Neodidymelliopsis, Aspergillus, Pseudopithomyces and Stemphylium. The yeasts Candida and Clavispora were particularly abundant in high-touch surfaces indoors., [Conclusions:] DNA metabarcoding revealed a much more comprehensive inventory of hospital fungi compared to culturing, however, both approaches found similar dominant taxa including a variety of potentially opportunistic human pathogens. DNA metabarcoding can assist hospital managers under certain demanding situations, e.g. construction works or reported microbial outbreaks, providing an in-depth characterization of hospital mycobiomes. In addition, qPCR proved to be a reliable method to quantify the fungal load in air samples, which can complement CFU and particle data in environmental assessments., Agencia Estatal de Investigación Unraveling the hospital mycobiomes associated with the invasive fungal infections and the emergence of antimicrobial resistancePID2021-123184OA-I, Peer reviewed

DATA FROM: HIGH THERMAL VARIATION IN MAXIMUM TEMPERATURES INVERT BRETT’S HEAT-INVARIANT RULE AT FINE SPATIAL SCALES

Discovering how species’ thermal limits evolve and vary spatially is crucial for predicting their vulnerability to ongoing environmental warming. Current evidence indicates that heat tolerance is less spatially variable than cold tolerance among species, presenting a major concern for organismal vulnerability in a rapidly warming world. This asymmetry in thermal limits has been supported by large-scale geographic studies, across latitudinal and elevational gradients (known as Brett´s heat invariant rule). Yet, how critical limits vary across finer spatial scales (e.g., across microenvironments) is less understood. Here, we show that minimum temperatures are more variable than maximum temperatures at large geographic scales (across latitude/elevation), but are less variable at local scales (within sites), guiding spatial asymmetries in thermal tolerances. Using thermal tolerance measurements from amphibians, insects, and reptiles, we confirm the invariance of heat tolerance at large spatial scales and also find more variable heat than cold tolerances at local scales (an inverted Brett’s heat invariant rule at fine spatial scales). Our results suggest that regional or global-level studies will likely obscure fine-scale structuring in thermal habitats and corresponding local heat tolerance adaptation patterns. We emphasize that inferences based on broadscale geographic patterns obscure fine-scale variation in thermal physiology. For instance, a genetic basis for fine-scale variation in thermal physiology may reshuffle spatial and phylogenetic patterns of vulnerability., CGL2017-86924-P, PID2022-140618NB-I00 2020AEP112_CSIC NSF DEB-2054569, Peer reviewed
