These data are associated to the paper with the same title and are structured according to the figures in the paper. These data correspond to the research results of the WP2 of the European Project 4D-POLYSENSE., The present dataset collects and reports the scientific results corresponding to a scientific study of a commercial and fully flexible versatile bilayer system, composed by a polypropylene (PP) mesh and a covalently bonded poly(N-isopropylacrylamide) (PNIPAAm) hydrogel. In this study the mechanism responsible for cell-adhesion and de-adhesion, when the PNIPAAm-co-MBA monolayer film (adherent on one hand to the PP substrate and anti-adherent on the other hand to cell proliferation) is being employed, has been elucidated. The grafting of the hydrogel was studied and the the graft yield as well as the spectroscopic characterization (FTIR and XPS) of samples treated at different concentration of the crosslinker and different grafting time are ported in the present dataset. The scientific results of cell adhesion and biocompatibility assay are also included, being the in vitro results with fibroblast (COS-1) and epithelial (MCF-7) cells obtained with a mesh modified with a porous iPP-g-PNIPAAm bilayer system, prepared via PNIPAAm grafting for 2 h at the lowest N,NI-methylene bis(acrylamide) (MBA) concentration (1 mM).

These data are associated to the paper with the same title and are structured according to the figures in the paper. These data correspond to the research results of the WP1 of the European Project 4D-POLYSENSE., In the scientific publication corresponding to the present dataset, a commercial and fully flexible monofilament polypropylene mesh has been used for the deposition of a thermosensitive hydrogel, generated by graft copolymerization of N-isopropylacrylamide (NIPAAm) and N,NI-methylene bis(acrylamide) (MBA) monomers. The study represents an initial step for the future development of a new generation of surgical meshes, able to interact with the complex three-dimensional structure of biological tissues, as smart materials working as self-evolving motion sensors. The meshes were previously functionalized by O2-plasma method and subsequently grafted with the thermoresponive hydrogel and the data related to micro- and standard spectroscopy techniques empoyed for the characterization, such as Raman spectroscopy, FTIR spectroscopy and XPS, are reported in the present dataset. Additionally, the data of graft yield and thermoresponsive properties of PNIPAAm (VPTT behaviour) are included in the dataset

These data are associated to the paper with the same title of the authors, and are structured according to the figures in the paper: http://hdl.handle.net/2117/335727, The use of Cold Atmospheric Plasma (CAP) in oncology is being recently studied as a novel potential anti-cancer therapy. However, the beneficial effects of CAP for treating osteosarcoma have mostly been demonstrated in 2-dimensional cultures of cells, which do not mimic the complexity of the 3-dimensional (3D) bone microenvironment. In order to evaluate the effects of CAP in a relevant context of the human disease, we developed a 3D tissue-engineered model of osteosarcoma using a bone-like scaffold made of collagen type I and hydroxyapatite nanoparticles. Human osteosarcoma cells cultured within the scaffold showed a high capacity to infiltrate and proliferate and to exhibit osteomimicry in vitro. As expected, we observed significantly different functional behaviors between monolayer and 3D cultures when treated with Cold Plasma-Activated Ringer’s Solution (PAR). Our data reveal that the 3D environment not only protects cells from PAR-induced lethality by scavenging and diminishing the amount of reactive oxygen and nitrogen species generated by CAP, but also favours the stemness phenotype of osteosarcoma cells. This is the first study that demonstrates the negative effect of PAR on cancer stem-like cell subpopulations in a 3D biomimetic model of cancer. These findings will allow to suitably re-focus research on plasma-based therapies in future

This dataset is related to the protocol developed by the authors on the evaluation of the effects of cold atmospheric plasma and plasma-treated liquids in cancer cell cultures. More specifically, the data correspond to the anticipated results presented in the paper submitted to Nature Protocols, The present dataset corresponds to the anticipated results that can be obtained following the protocol described in the article. In particular, dataset corresponding to Figure 8 presents the absorbance values from the WST-1 cytotoxicity assay as a function of the well size where the experiment is performed. Data from Figure 9b show absorbance values from the WST-1 cytotoxicity assay as a function of the treatment time. Data from figure 9c-e show the absorbance or fluorescence values obtained in the experiments to quantify RONS, together with the values from the calibration curves. Data fromfigure 10 a provide the list and values of mRNA fold induction of the genes analyzed, and data from figure 10c show the pixel intensity obtained for the phospho-kinase antibody array.

INFERRING SEDIMENT TRANSFERS AND FUNCTIONAL CONNECTIVITY OF RIVERS FROM REPEAT TOPOGRAPHIC SURVEYS (SUPPORTING INFORMATION)

  • Calle, Mikel
  • Calle, Javier
  • Alho, Petteri
  • Benito, Gerardo
High-resolution topographic models have revolutionized monitoring of river changes by comparing sequential river topographic surveys (i.e. change detection). Nevertheless, much more may be obtained from this innovative quantification of changes. In this paper, we enhance the interpretation of geomorphic processes by presenting a new method for understanding of sources and sinks of sediment, river sediment transfers and functional sediment connectivity. Repeat digital elevation models (DEMs) obtained by photogrammetry were used to quantify topographic change after two floods by creating a DEM of difference (DoD) of a 6.5 km-long reach of Rambla de la Viuda stream, an ephemeral gravel-bed river in eastern Spain. The proposed method involved dividing the channel into 10 m-long longitudinal strips that were used to systematically draw boundaries between the erosional and depositional areas of the DoD. The analysis objectively: (i) drew a series of erosional and depositional segments, from 120 to 1360 m in length; (ii) estimated ranges of source-to-storage sediment transport distances, 320–670 m in the upstream and middle reaches and up to 2030 m in the lower reach; and (iii) obtained values of functional connectivity (i.e. the ratio between the sediment exported (erosion) and retained (deposition), ranging from 103 to 10−3). The variability in these three parameters along the river was found to be related to the level of channel disturbance by in-stream mining during the 1990s and 2000s. Additionally, this method indicates that the main process responsible for self-adjustment of the present morphosedimentary conditions is intra-reach erosion of banks and channel beds. Thus, this study proposes a new methodology to characterize morphological change, sediment transfer and connectivity that may serve as environmental indicators of the hydromorphological integrity of rivers with potential application to the European Water Framework Directive.
